• Find and separate genetic material from the sample
• Tests for RNA sequence
• Rapidly develop a qualitative PCR detection w/o purifying the virus

He also said, "The number of cycles is usually carried out 25-35 times but may vary upon the amount of DNA input and the desired yield of PCR product. If the DNA input is fewer than 10 copies, up to 40 cycles may be required to produce a sufficient yield."

Since the first cluster of cases was identified in Wuhan City, China, in December, 2019, coronavirus disease 2019 (COVID-19) has rapidly spread across China, causing multiple introductions in 109 countries/territories/areas as of March 10th. Despite the scarcity of publicly available data, scientists around the world have made strides in estimating the magnitude of the epidemic, the basic reproduction number, and transmission patterns. Recently more evidence suggests that a substantial fraction of the infected individuals with the novel coronavirus show little if any symptoms, which suggest the need to reassess the transmission potential of this emerging disease.

In this study, we derive estimates of the transmissibility and virulence of COVID-19 in Wuhan City, China, by reconstructing the underlying transmission dynamics using multiple data sources.

Methods: We employ statistical methods and publicly available epidemiological datasets to jointly derive estimates of transmissibility and severity associated with the novel coronavirus. For this purpose, the daily series of laboratory-confirmed COVID-19 cases and deaths in Wuhan City and epidemiological data of Japanese evacuees from Wuhan City on board government-chartered flights were integrated into our analysis.

Results: Our posterior estimates of basic reproduction number (R) in Wuhan City, China in 2019-2020 reached values as high as 5.20 (95%CrI: 5.04-5.47) and the enhanced public health intervention after January 23rd in 2020 was associated with a declined R at 0.58 (95%CrI: 0.51-0.64), with the total number of infections (i.e. cumulative infections) estimated at 1905526 (95%CrI: 1350283-2655936) in Wuhan City, raising the proportion of infected individuals to 19.1% (95%CrI: 13.5-26.6%). We also found that most recent crude infection fatality ratio (IFR) and time-delay adjusted IFR is estimated to be 0.04% (95% CrI: 0.03-0.06%) and 0.12% (95%CrI: 0.08-0.17%), which is several orders of magnitude smaller than the crude CFR estimated at 4.19%.

Conclusions: We have estimated key epidemiological parameters of the transmissibility and virulence of COVID-19 in Wuhan, China during January-February, 2020 using an ecological modelling approach. The power of our approach lies in the ability to infer epidemiological parameters with quantified uncertainty from partial observations collected by surveillance systems.

Yet here are some further notes that show even clearer evidence that the pandemic is overblown.

• The website of Dr. Wolfgang Wodarg, one of the earliest and internationally best known critics of the Covid19 panic, was deleted for a few hours today by the German provider Jimdo and only went online again after strong protests. It is not known whether the temporary deletion was due to general complaints or a political instruction.
  
  
• The university email address of emeritus professor Dr. Sucharit Bhakdi, who wrote an Open Letter to Chancellor Angela Merkel, was deactivated earlier, but was also reactivated after protests.
  
  
• The Swedish government has announced that it is going to officially distinguish between deaths "by“ and deaths "with“ the coronavirus, which should lead to a reduction in reported deaths. Meanwhile, for some reason, international pressure on Sweden to abandon its liberal strategy is steadily increasing.
  
  
• The Hamburg health authority now has test-positive deaths examined by forensic medicine in order to count only "real" corona deaths. As a result, the number of deaths has already been reduced by up to 50% compared to the official figures of the Robert Koch Institute.
  
• The British project "In Proportion" tracks mortality "with" Covid19 in comparison to influenza mortality and all-cause mortality, which in Great Britain is still in the normal range or below and is currently decreasing.

All of that leads us back to the infamous Diamond Princess, if Syd_M's tiny brain actually can hold out memory that long. 700 infected passengers and crew. Only 7 died all of which are senior citizens. Japan to declare nationwide state of emergency as virus spreads

Prime Minister Shinzo Abe has decided to declare a nationwide state of emergency in an attempt to prevent the further spread of the new coronavirus, a government official said Thursday.

The government had been considering expanding of the state of emergency, currently covering Tokyo, Osaka and five prefectures, to also include Kyoto, Aichi and Hokkaido. But instead it will likely be extended nationwide due to a rise in infections in other parts of Japan.

Well, would you look at that. So much for your assertion that it's all a hoax because Japan didn't quarantine. Only an idiot would not have realized that Japan resisted taking stronger measures because they didn't want to jeopardize the Olympics, which they were to host. It wasn't until late March when it was officially announced that the Olympics would be postponed did Japan start taking it seriously. And now they'll be entering a state of emergency. They started very late, but now it's moving, and it will keep on moving because a virus doesn't care about your conspiracy theories.

At some point, lockdown as strategy will have to be weighed against the risk of economic collapse and social unrest. The media cannot continue to condemn Hydroxycholoroquine as "snake oil" just because Trump said it was promising. The cases don't lie. Doctor Wang in Cebu recovered as a result of the combination of Hydroxycholoroquine and Azythromycin (some say adding Zinc into the mix would be the best), as attested to by Dr. Larazabal in the local news. The problem here is most doctors who need to follow DOH/WHO guidelines cannot recommend this drug regimen to patients. The only way for a patient to get these is to perhaps sign a waiver to clear the hospital of any liability.
