Managerless Aston Villa host West Ham United who will be playing their first competitive match under Avram Grant.
Wolverhampton Wanderers have something of a local derby against Stoke City, Everton go to Blackburn Rovers and Sunderland manager Steve Bruce meets his former club Birmingham City at the Stadium of Light.
The opening day's programme finishes with champions Chelsea opening the defence of their crown at home to newly-promoted West Bromwich Albion.
Spurs v Man City
This is an early re-run of last season's intriguing race for a Champions League place which saw Spurs edge out City in the final week of the campaign.
Harry Redknapp's men made a declaration of their intent by beating Liverpool in their opening fixture of last season and will be keen to serve notice of a repeat.
In a bid to redress that balance of power, City have spent around £75m this summer with a promise of more to follow. By contrast the north London club have brought in just one new face.
Spurs have won 11 of the last 12 Premier League meetings between the clubs but have won just two of their last nine opening fixtures.
Aston Villa v West Ham
Aston Villa face an uncertain start to the season following the resignation of manager Martin O'Neill.
Reserve team coach Kevin MacDonald has taken temporary charge as Villa face opposition who are also under new management with Avram Grant now at the helm.
Villa have won only one of their last five opening-day fixtures and will be hoping to get off to a better start than they did last season when they lost at home to Wigan.
The Hammers have won four of their last five first-day fixtures but have won only one of their last 13 visits to Villa Park.
Blackburn v Everton
Rovers have finished in the top half in four of the last five seasons, thanks largely to their strong home form.
Everton were one of only three visiting sides to win at Ewood Park last season and Blackburn will be keen to avenge April's 3-2 defeat which was their only reverse in their last nine fixtures.
The Merseysiders are unbeaten in their last six visits to Blackburn, although four of those matches have been drawn.
Everton are unbeaten in 11 Barclays Premier League matches since losing 2-1 at Tottenham in February, their longest unbeaten run since 1986.
Bolton v Fulham
Mark Hughes faces a tough test as he takes charge of Fulham for the first time in a competitive match, having guided them to an impressive 5-1 win over Werder Bremen in his only outing to date.
One of his main priorities will be to improve the club's away form with only seven wins on the road in the last three seasons. Only one of those came last season and that was on the opening day, a 1-0 success at Portsmouth.
Fulham have won only one of their last six visits to the Reebok but Bolton have not beaten the Cottagers since February 2007.
This is the fifth successive season that the Trotters have kicked off in front of their own fans. Fulham have scored one goal in each of their last four opening-day fixtures.
Sunderland v Birmingham
The Black Cats have brought in eight new faces over the summer, including some canny loan signings in Nedum Onuoah and Danny Welbeck from the two Manchester clubs.
Birmingham, who scored just three headers last season, will look to the height of six foot eight inch striker Nikola Zigic.
The Blues have won five of their eight Premier League meetings with the Black Cats who will again look to the firepower of 25-goal striker Darren Bent.
Sunderland have managed just five goals in their nine opening-day fixtures in the Premier League.
Wigan v Blackpool
Blackpool are relishing their first appearance in the Premier League as they return to the top flight after a 40-year absence.
Under the charge of charismatic manager Ian Holloway, the Seasiders are eager to make their mark in a fixture which has been reversed. Blackpool were originally due to be at home but ground improvements prompted a switch to the DW Stadium.
All eyes will be on the man in the middle as referee Mark Halsey makes an emotional return to the Premier League following a year-long battle against cancer.
Wolves v Stoke
Wolves finished strongly in 15th place last season while Stoke ended up 11th, one place higher than in their debut Barclays Premier League campaign the previous year.
Both will again make survival their immediate priority and have strengthened significantly over the summer with a view to achieving that aim.
Stoke could give a debut to new club record signing Kenwyne Jones while Wanderers have added Steven Fletcher, Stephen Hunt and Jelle van Damme.
Last season, both meetings between the clubs ended level. Wolves have conceded a total of seven goals in their two previous opening Premier League fixtures and Stoke will hope to maintain last season's improvement on the road which saw them double the 10 away points they picked up in 2008/09.
Chelsea v West Brom
Newly-promoted West Brom could hardly have had a tougher start to life back in the Barclays Premier League as they travel to defending champions Chelsea. Their next three away matches are Liverpool, Arsenal and Manchester United.
The Blues finished last season in spectacular form at Stamford Bridge scoring 23 goals in their final four fixtures in front of their own fans as they set a new Premier League record of 103 goals scored in a season.
Didier Drogba scored 29 goals in 32 matches to claim the Barclays Golden Boot and Albion will look to the experience of new signing Pablo Ibanez.
Chelsea will be looking to win their opening match for the ninth successive season.
Albion have lost all eight previous Premier League meetings with Chelsea, scoring just twice.
